Kajian Jenis Bahan Penstabil dan Metode Pembuatan pada Pengembangan Sediaan Nanosuspensi untuk Penghantaran Agen Sitotoksik

Abstract


ABSTRACT: Cancer is a disease that originates from the body's tissue cells which is characterized by rapid cell division. One form of nanoparticle delivery that is widely used for cancer treatment with the aim of increasing the bioavailability of higher drugs is nanosuspension. The success of the development of nanosuspension preparations is influenced by two things, namely the type of stabilizer and the method of manufacture. This study aims to determine the effect of the type of stabilizer and the method of manufacture in the development of nanosuspension preparations on the activity of cytotoxic agents. This research was conducted using a Systematic Literature Review (SLR) based on inclusion and exclusion criteria. The results of the study found 11 active compounds that have been developed into nanosuspension preparations including deacety mycoepoxydiene, celastrol, annonaceous acetogenins, curcumin, oridonin, hydroxy-genkwanin, genkwanin, ceramide and docetaxel, dilsufiram, and PIK-75. Stabilizers that can be used for the development of nanosuspension preparations are polymers or surfactants, namely lecithin, poloxamer-188, HPMC, PVP, sodium deoxycholate (SDC), TPGs (D-α-Tocopheryl polyethylene glycol succinate 1000), and hydroxypropyl-beta- cyclodextrin (HP-β-CD), where this type of stabilizer is able to stabilize nanosuspension by steric, electrostatic, and electrosteric stabilization and nanosuspension preparations can be developed using top-down or bottom-up methods.

ABSTRAK: Kanker merupakan penyakit yang berasal dari sel-sel jaringan tubuh yang ditandai dengan pembelahan sel yang cepat. Salah satu bentuk penghantaran nanopartikel yang banyak digunakan untuk pengobatan kanker dengan tujuan untuk meningkatkan bioavaibilitas obat yang lebih tinggi adalah nanosuspensi. Keberhasilan pengembangan sediaan nanosuspensi dipengaruhi oleh dua hal yaitu jenis bahan penstabil dan metode pembuatannya. Kajian ini bertujuan untuk mengetahui pengaruh jenis bahan penstabil dan metode pembuatan dalam pengembangan sediaan nanosuspensi terhadap aktivitas agen sitotoksik. Penelitian ini dilakukan secara Systematic Literature Review (SLR) dengan berdasarkan kriteria inklusi dan eksklusi. Hasil kajian mendapatkan 11 senyawa aktif yang telah dikembangkan menjadi sediaan nanosuspensi diantaranya adalah deacety mycoepoxydiene, celastrol, annonaceous acetogenins, curcumin, oridonin, hidroksi-genkwanin, genkwanin, ceramide dan docetaxel, dilsufiram, dan PIK-75. Bahan penstabil yang dapat digunakan untuk pengembangan sediaan nanosuspensi yaitu seperti golongan polimer atau surfaktan yaitu lesitin, poloksamer-188, HPMC, PVP, sodium deoxycholate (SDC), TPGs (D-α-Tocopheryl polyethylene glycol succinate 1000), dan hidroksipropil-beta-siklodekstrin (HP-β-CD), dimana jenis bahan penstabil tersebut mampu menstabilkan nanosuspensi secara stabilisasi sterik, elektrostatik, dan elektrosterik serta sediaan nanosuspensi dapat dikembangkan dengan menggunakan metode top-down atau bottom-up.
