Peningkatan Stabilitas Asam dari Lansoprazol dengan Teknik Kokristalisasi Menggunakan Koformer Natrium Karbonat

Abstract


Abstract. Lansoprazole (LAN) is a gastric ulcer drug from the class of proton pump inhibitors that have unstable properties in acidic conditions, so that the effectiveness of the drug becomes low when in acidic conditions. In this research has been done co-crystallization technique LAN using a co-former sodium carbonate (NK) with a grinding method (neat grinding and solvent-drop grinding) which aims to see the interactions between LAN with NK and see the results of stability acid tests on cocrystal LAN -NK. The solid co-crystallization result were characterized, which include thermal analysis using differential scanning calorimetry (DSC), crystallographic analysis using powder x-ray diffraction (PXRD) and morphological analysis using a scanning electron microscope (SEM). The results showed that the molecular interactions is not formed between LAN-NK, but NK just dispersed in the LAN at a mole ratio of 1:1. Furthermore, the acid stability test using high performance liquid chromatography (HPLC) indicated that a slight improve in acid stability LAN of co-crystallization results.

Abstrak. Lansoprazol (LAN) merupakan salah satu obat tukak lambung dari golongan proton pump inhibitor yang memiliki sifat tidak stabil terhadap suasana asam, sehingga efektivitas obat menjadi rendah ketika berada dalam suasana asam. Pada penelitian ini telah dilakukan teknik kokristalisasi LAN menggunakan koformer natrium karbonat (NK) dengan metode penggilingan (neat grinding dan solvent drop grinding) yang bertujuan untuk melihat interaksi yang terjadi antara LAN dengan NK dan melihat hasil pengujian stabilitas asam pada kokristal LAN-NK. Padatan hasil kokristalisasi dikarakterisasi, yang meliputi analisis termal menggunakan differential scanning calorimetry (DSC), analisis kristalografi menggunakan powder x-ray diffraction (PXRD) dan analisis morfologi menggunakan scanning electron microscope (SEM). Hasil karakterisasi menunjukkan bahwa tidak terbentuk interaksi molekular antara LAN-NK, melainkan NK hanya terdispersi dalam LAN pada perbandingan mol 1:1. Selanjutnya pengujian stabilitas asam menggunakan kromatografi cair kinerja tinggi (KCKT) menunjukkan bahwa terlihat adanya sedikit peningkatan stabilitas asam pada LAN hasil kokristalisasi.
