Pengembangan Metode Deteksi Residu Bahan Kimia Obat dalam Daging Sapi dari Perdagangan dengan Cara Kromatografi Lapis Tipis dan Kromatografi Cair Kinerja Tinggi

Abstract


Abstract. The study has developed analysis method of drugs (BKO) salbutamol, chloramphenicol, spiramycin and tetracycline on beef using solid phase extraction methods, thin layer chromatography (TLC), high performance liquid chromatography (HPLC). The research phase includes the preparation of samples of beef by centrifugation using a solution of trichloroacetic acid (TCA) 20% which will obtain supernatant. The result of supernatant is extracted in solid phase using a solvent extraction of ethanol, distilled water washing solution, and a solution of oxalic methanol elution. Extraction which is obtained in optimization by TLC using acetonitrile motion phase: methanol (1: 4) (v / v) showed the most excellent optimization in attracting salbutamol, chloramphenicol, spiramycin and tetracycline. Optimization is continued using stationary phase HPLC using a C-18 motion phasemethanol:water containing 0.1% acetic acid (75:25) (v / v) generate most excellent analysis on salbutamol and chloramphenicol with the type of gradient elution and UV detector 280 nm.

Abstrak. Telah dilakukan pengembangan metode analisis residu bahan kimia obat (BKO) salbutamol, kloramfenikol, spiramisin, dan tetrasiklin pada daging sapi menggunakan metode ekstraksi fasa padat, kromatografi lapis tipis (KLT), kromatografi cair kinerja tinggi (KCKT). Tahap penelitian meliputi preparasi sampel daging sapi dengan sentrifuga menggunakan larutan trichloroacetic acid (TCA) 20% yang nantinya akan mengahasilkan supernatan. Supernatan yang dihasilkan  diekstraksi fasa padat menggunakan pelarut ekstraksi metanol, larutan mencuci akuades, dan larutan pengelusi metanol oksalat. Ekstraksi yang didaptkan di optimasi dengan KLT menggunakan fase gerak asetonitril:metanol (1:4) (v/v) menunjukan optimasi paling baik dalam menarik salbutamol, kloramfenikol, spiramisin, dan tetrasiklin. Dilanjutkan optimasi denagn KCKT menggunakan fase diam C-18 fase gerak metanol:air yang mengandung asam asetat 0,1% (75:25) (v/v) menghasilkan analisa paling baik pada salbutamol dan kloramfenikol dengan tipe elusi gradien dan detektor UV 280 nm.
