Efek Samping Ekstrak Air Buah Pepaya Muda (Carica papaya L.) sebagai Laktagogum terhadap Escherichia Coli Secara In Vitro

Abstract


Abstract: Papaya as herbal medicines have pharmacological benefits in every part. Earlier research found that water extract of papaya fruit unripped has an effect as facilitator of breast milk at dose of 20mg/mL. Papaya also known to have an antibacterial effect against Escherichia coli at certain doses. Active antibacterial substance that contained in papaya fruits are alkaloids, flavonoids, tannins and triterpenoids. This study aims to assess the antibacterial effect from inhibition zone method water extract of papaya fruit unripped to Escherichia coli as normal flora at lactagogum dose This research was conducted using pure laboratory experimental in vitro method. Samples derived from pure culture of Escherichia coli which obtained from the Laboratory STIKES UNJANI. Test material is dose of extract water of papaya fruits unripped 20mg/mL, 30ug chloramphenicol as a positive control and Tween 80 as a negative control. Antibacterial test was conducted using a modified diffusion Kirby bauer with Mueler-Hilton agar were performed in 9 repetition. Antibacterial test results showed the formation of inhibition zone at doses of 20mg/mL and chloramphenicol as positive control. Mann-Whitney statistic test showed meaningful result. The conclusion of this study is the water extract of papaya fruit unripped has antibacterial side effect against Escherichia coli.

Abstrak: Pepaya sebagai obat herbal memiliki manfaat farmakologis pada setiap bagiannya. Penelitian sebelunya ditemukan bahwa ekstrak air buah papaya muda memiliki efek sebagai pelancar ASI pada dosis 20mg/mL. Pepaya diketahui juga memiliki efek antibakteri terhadap Escherichia coli pada dosis tertentu. Zat aktif yang memiliki efek antibakteri terkandung dalam buah pepaya yaitu alkaloid, flavonoid, tannin dan triterpenoid. Penelitian ini bertujuan untuk menilai efek samping antibakteri ekstrak air buah papaya muda melalui pengukuran zona hambat terhadap flora normal Escherichia coli pada dosis buah papaya sebagai laktagogum. Penelitian ini dilakukan menggunakan metode eksperimental laboratorik secara in vitro. Sampel berasal dari biakan murni Escherichia coli yang diperoleh dari Laboratorium STIKES UNJANI. Bahan uji berupa ekstrak air buah papaya muda dengan dosis 20 mg/mL, kloramfenikol 30 µg sebagai kontrol positif dan Tween 80 sebagai kontrol negatif. Uji antibakteri dilakukan dengan metode difusi modifikasi Kirby bauer dengan medium agar Mueler Hilton yang dilakukan sebanyak 9 kali pengulangan. Hasil uji antibakteri menunjukan terbentuknya zona hambat, pada dosis 20 mg/mL dan kloramfenikol sebagai kontrol positif. Hasil uji statistik menggunakan Mann-Whitney menunjukan hasil yang bermakna. Kesimpulan dari penelitian ini adalah ekstrak air buah papaya muda memiliki efek samping antibakteri terhadap Escherichia coli.
