Penghambatan Escherichia coli ATCC 25922 oleh Fraksi Etil Asetat Daun Sirsak (Annona muricata Linn)

Abstract


Abstract: Escherichia coli is a normal flora in the digestive tract of humans and animals, and aerobic organisms residing in the gut main. Some strains of Escherichia coli develop resistance to several classes of antibiotics such as beta-lactam, quinolones, and aminoglycosides. Empirically, leaves of soursop (Annona muricata Linn) used for herbal medicine to treat various diseases. Compounds tannins, alcaloids, steroids, and flavonoids that containded in ethyl acetate fraction of soursop leaves an active substance that has antibacterial effect. This study aims to assess content of active substance in ethyl acetate fraction of soursop leaf and assess inhibition effect against Escherichia coli ATCC 25922. The study was conducted with pure experimental methods such as in vitro. The sampels are pure cultures Escherichia coli ATCC 25922 were obtained from laboratory of STIKES. The material test of ethyl acetate fraction of leaves of soursop (Annona muricata Linn) with a concentration of 20%, 40%, 60%, 80%, tween 80 as negative control, and chloramphenicol as positive control. Antibacterial effect against Escherichia coli ATCC 25922 tested using Kirby-bauer method on eosin methylen blue agar media by measuring inhibition zone, performed four times. The data in test for normality using Shapiro-wilk test and then continued by one way anova. Antibacterial test results showed inhibition zone formation at a concentration of 80% with an average diameter of 7 mm. Intergroup comparison with negative control and concentration of 80% showed a significant difference with p value is 0.000 (p value<0.05). The conclusion of this study is ethyl acetate fraction has an inhibition effect, but with a very minimal level.

Abstrak: Escherichia coli merupakan flora normal di dalam saluran pencernaan manusia dan hewan, dan organisme aerobik utama yang berada di usus. Beberapa strain Escherichia coli mengalami resistensi terhadap beberapa golongan antibiotik seperti golongan beta laktam, kuinolon, dan aminoglikosid. Secara empiris, daun sirsak (Annona muricata Linn) digunakan sebagai obat herbal untuk mengatasi berbagai penyakit. Senyawa tannin, alkaloid, steroid, dan flavonoid yang terkandung dalam fraksi etil asetat daun sirsak merupakan zat aktif yang memilki efek antibakteri. Penelitian ini bertujuan untuk mengetahui dan menilai zona hambat fraksi etil asetat terhadap Escherichia coli ATCC 25922. Penelitian dilakukan dengan metode eksperimental murni in vitro, sampel berasal dari biakan murni Escherichia coli ATCC 25922 yang diperoleh dari laboratorium STIKES jendral ahmad yani. Bahan uji berupa fraksi etil asetat daun sirsak (Annona muricata Linn) dengan konsentrasi 20%, 40%, 60%, 80%, kontrol negatif berupa tween 80, dan klorampenikol sebagai kontrol positif. Efek fraksi etil asetat terhadap Escherichia coli ATCC 25922 diuji dengan menggunakan metode Kirby-Bauer pada media eosin methylen blue agar dengan mengukur diameter zona hambat dan dilakukan sebanyak empat kali. Data di uji normalitas menggunakan uji Shapiro-Wilk lalu dilanjutkan dengan uji One Way Anova. Hasil uji antibakteri menunjukkan terbentuknya zona hambat pada konsentrasi 80% dengan diameter rata-rata 7 mm. Perbandingan antarkelompok kontrol negatif dengan konsentrasi 80% menunjukan adanya perbedaan bermakna dengan nilai p 0,000 (nilai p < 0,05). Kesimpulan dari penelitian ini adalah fraksi etil asetat memiliki efek zona hambat namun dengan kadar yang sangat minimal.
