Penurunan Efek Hemolisis Streptococcus pyogenes oleh Fraksi Etil Asetat Daun Sirsak (Annona muricata Linn)
Abstract
Abstrack: Streptococcus pyogenes is bacteria cause various diseases such as pharyngitis and many diseases. Antibiotics such as tetracycline and eritromycin have a high resistance rates. Empirically, soursop leaf (Anonna muricata Linn) has been widely use as herbal for treating diseases. Active compunds such as annonaceous acetogenin, alkaloid, flavonoid, and steroid contain effectiveness antibacterial effect. Importance  this study to assess effectiveness antihemolysis effect of ethyl acetate fraction soursop leaves to Streptococcus pyogenes. The study was conducted with pure experimental in vitro, samples are Streptococcus pyogenes. Test material are ethyl acetate fraction soursop leaves at concentration 20%, 40%, 60% and 80% with negative and positive control are Ttrypticase soy blood agar wich cultured by Streptococcus pyogenes without ethyl acetate faction soursop leaves and Ttrypticase soy blood agar without Streptococcus pyogenes. Hemolysis effect tested by see color changes on TSA blood agar. The results of hemolysis test showed decrease wich given fraction ethyl acetate soursop leaf with a concentration of 20%, 40%, 60%, and 80%. Conclusion is ethyl acetate fraction soursop leaves can reduce the effect of hemolysis Streptococcus pyogenes.Â
Abstrak: Streptococcus pyogenes merupakan bakteri yang dapat menyebabkan berbagai macam penyakit seperti faringitis dan penyakit lainya. Berapa antibiotik untuk bakteri ini seperti tetrasiklin dan erytromisin memiliki angka resistensi yang tinggi. Secara empiris, daun sirsak (Annona muricata Linn) banyak digunakan sebagai obat herbal untuk menanggulangi berbagai penyakit. Senyawa aktif seperti annoneceus acetogenin, tanin, alkaloid, flavonoid, steroid yang terkandung memiliki efek antibakteri. Penelitian ini bertujuan untuk menilai efek penurunan hemolisis fraksi etil asetat daun sirsak terhadap Streptococcus pyogenes. Penelitian dilakukan dengan metode eksperimental murni in vitro, sampel berasal dari biakan Streptococcus pyogenes. Bahan uji berupa fraksi etil asetat daun sirsak pada konsentrasi 20%, 40%, 60%, 80% dengan kontrol negatif dan positif yaitu Trypticase soy blood agar  yang dikultur dengan Streptococcus pyogenes tandpa diberikan fraksi etil asetat dan Trypticase soy blood agar  tanpa dikultur dengan Streptococcus Pyogenes. Pengujian efek hemolisis dinilai dengan melihat perubahan warna yang terjadi pada TSA blood agar. Hasil uji hemolisis menunjukkan adanya penurunan hemolisis pada Streptococcus pyogenes yang telah diberikan fraksi etil asetat daun sirsak dengan konsentrasi 20%, 40%, 60%, dan 80%. Kesimpulan penelitian ini adalah fraksi etil asetat daun sirsak dapat menurunkan efek hemolisis Streptococcus Pyogenes.
