Daya Antibakteri Ekstrak Etanol Daun Pepaya (Carica Papaya Linn) terhadap Streptococcus Pyogenes

Abstract


Abstract. Indonesian people have long used plants as traditional medicines for healing bacterial infections. One of the plants that can be used as traditional medicine is papaya. However, research data and adequate research methodologies for evaluating the effects of traditional medicines are still lacking. Streptococcus pyogenes became one of the bacteria that causes infection that the incidence rate is still high in Indonesia. The aim of this research is to know the antibacterial power of papaya leaf ethanol extract (Carica papaya Linn) to Streptococcus pyogenes bacteria and assess the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of papaya leaf ethanol extract against Streptococcus pyogenes bacteria. This research was conducted by laboratory descriptive methods. Antibacterial tests were performed by diffusion and dilution methods with streak confirmation. The research was conducted in Microbiology Laboratory of POLTEKKES Bandung in September-November 2018 with the extraction process of papaya leaf. The result of antibacterial test by diffusion method showed the formation of inhibition zone at concentration of 100% ethanol extract of papaya leaf with an average diameter of 16,37 mm and were bacteriostatic, because the clear zone formed with bacterial growth. The results of the dilution method showed that MIC in the ethanol extract of papaya leaves at concentration 25%, whereas for MBC it could not be determined. Active substances such as flavonoids, tannins, saponins, papaim and karpain act as an antibacterial agent. This study showed that there is an antibacterial power of ethanol extract of papaya leaf against Streptococcus pyogenes bacteria.

Abstrak. Masyarakat Indonesia telah lama menggunakan tanaman sebagai obat tradisional bagi penyembuhan penyakit infeksi bakteri. Salah satu tanaman yang dapat dimanfaatkan sebagai obat tradisional adalah tanaman pepaya. Namun data penelitian serta metodologi penelitian yang memadai untuk mengevaluasi efek obat tradisional masih sangat kurang. Streptococcus pyogenes menjadi salah satu bakteri penyebab infeksi yang angka kejadiannya masih tinggi di Indonesia. Penelitian ini bertujuan untuk mengetahui daya antibakteri ekstrak etanol daun pepaya (Carica papaya Linn) terhadap bakteri Streptococcus pyogenes dan menilai konsentrasi hambat minimal serta konsentrasi bunuh minimal ekstrak etanol daun ekstrak etanol daun pepaya (Carica papaya Linn) terhadap bakteri Streptococcus pyogenes. Penelitian ini dilakukan dengan metode deskriptif laboratorik. Uji antibakteri dilakukan dengan metode difusi dan dilusi dengan konfirmasi streak. Penelitian dilakukan di Laboratorium Mikrobiologi POLTEKKES Bandung pada bulan September-November 2018 bersamaan dengan proses ekstraksi daun pepaya. Hasil uji antibakteri dengan metode difusi menunjukan terbentuknya zona hambat pada konsentrasi 100% ektrak etanol daun pepaya (Carica papaya Linn) dengan diameter rata-rata 16,37 mm dan bersifat bakteriostatik, hal ini dikarenakan pada area tersebut terbentuk zona bening yang terdapat pertumbuhan bakteri. Hasil uji dengan metode dilusi ditemukan KHM pada ekstrak etanol daun pepaya konsentrasi 25%, sedangkan untuk KBM tidak dapat ditentukan. Zat aktifnya seperti flavonoid, tanin, saponin, papaim dan karpain diperkirakan berperan dalam efek antibakteri. Penelitian ini menunjukkan bahwa terdapat daya antibakteri ekstrak etanol daun pepaya (Carica papaya Linn)  terhadap bakteri Streptococcus pyogenes.
