Pengujian Aktivitas Antibakteri Minuman Kefir terhadap Bakteri Escherichia coli dan Staphylococcus aureus

Abstract. The testing for antibacterial activity in kefir drink used three samples of kefir milk with 10%, 50%, 70%, 90% concentration , and undiluted kefir. The sample were tested for the pH and antibacterial activity againts Escherichia coli and Staphylococcus aureus in each kefir. The antibacterial activity consists of several stages; testing the purity and identification, determining the diameter of the obstruction, the bactericidal degree, testing the potential of kefir as antibactery compared to the antibiotic chloramphenicol on Staphylococcus aureus as well as processing the data in accordance with 95% ANOVA level confidence.The result of data showed that there are no significant differences between the three samples of kefir drink, but the test result of antibacterial activity in kefir B sample has an activity which on the bacteria Escherichia coli 1 mL of kefir B sample with 100% concentration is equivalent to 2,093 g (0,002039 mg) chloramphenicol while for Staphylococcus aureus 1 mL of kefir B sample with 100% concentration is equivalent to 0,209 g (0,000209 mg) of tetrasiklin.

Abstrak. Pengujian aktivitas antibakteri minuman kefir menggunakan tiga sampel kefir susu sapi dengan konsentrasi 10%, 30%, 50%, 70%, 90% dan kefir tanpa pengenceran. Sampel minuman kefir dilakukan pengujian pH tiap kefir dan uji aktivitas antibakteri terhadap bakteri Escherichia coli dan Staphylococcus aureus. Aktivitas antibakteri terdiri dari beberapa tahap yaitu uji kemurnian dan identifikasi, penentuan diameter hambat, penentuan konsentrasi hambat minimum, penentuan kadar bakterisid, pengujian potensi minuman kefir sebagai antibakteri yang dibandingkan terhadap antibiotik kloramfenikol untuk Escherichia coli dan antibiotik tetrasiklin untuk Staphylococcus aureus serta pengolahan data secara ANOVA dengan taraf kepercayaan 95%.  Hasil yang diperoleh tidak ada perbedaan bermakna antara ketiga sampel minuman kefir, tetapi dari hasil pengujian aktivitas antibaketri sampel minuman kefir B memiliki aktivitas antibakteri dimana untuk bakteri Escherichia coli 1 mL sampel kefir B konsentrasi 100% ekuivalen dengan 2,039 mg (0,002039 mg) kloramfenikol sedangkan untuk Staphylococcus aureus 1 mL sampel kefir B konsentrasi 100% ekuivalen dengan 0,209 mg (0,000209 mg) tetrasiklin.
