Peningkatan Stabilitas Asam dari Omeprazol dengan Teknik Kokristalisasi Menggunakan Koformer Natrium Karbonat

Abstract. Omeprazole (OMEP) is one of the benzimidazole class of drugs which has selective proton pump inhibitory activity. OMEP has a low stability to acidic conditions in the stomach. In this study has been carried out co-crystallization of omeprazol used sodium carbonate (NK) as coformer with neat grinding and solvent drop grinding methods, which aims to see interaction between OMEP and NK and also see the test results of co-crystallization OMEP-NK on the stability of the acid. The solid phase was characterized by using differential scanning calorimetry, powder x-ray diffraction and scanning electron microscope. The identification and characterization result showed that no formation of molecular compounds between OMEP-NK at the mole ratio of 1:1. Furthermore, the acid stability testing using HPLC method. The acid stability of the co-crystallization product showed a slight improve by using HPLC test.

Abstrak. Omeprazol (OMEP) merupakan salah satu obat golongan benzimidazol dengan aktivitas penghambatan pompa proton selektif. OMEP memiliki kestabilan yang rendah terhadap kondisi asam di lambung. Pada penelitian ini telah dilakukan kokristalisasi OMEP menggunakan koformer natrium karbonat (NK) dengan metode neat grinding dan solvent drop grinding yang bertujuan untuk melihat interaksi yang terjadi antara OMEP dengan NK dan melihat hasil pengujian kokristalisasi OMEP-NK terhadap stabilitas asam. Padatan hasil kokristalisasi ditentukan karakteristik fisikanya dengan menggunakan differential scanning calorimetry, powder x-ray diffraction dan scanning electron microscope. Hasil identifikasi dan karakterisasi menunjukkan tidak terbentuknya interaksi molekular antara OMEP-NK pada perbandingan mol 1:1. Selanjutnya, dilakukan pengujian stabilitas asam menggunakan metode KCKT. Terlihat adanya sedikit peningkatan stabilitas asam pada OMEP hasil kokristalisasi.
