Efek Preventif Ekstrak Etanol Buah Mengkudu (Morinda Citrifolia L) terhadap Penurunan Jumlah Eritrosit Tikus yang Diinduksi Metotreksat

Abstract

Abstract: Noni fruit (Morinda citrifolia) is a plant that has long been used as a treatment, one alternative treatment for anemia that is by increasing the number of erythrocytes. This is possible because of terpenoid contained in noni fruit has the function to help recovery of body’s cell. This study is aimed to know the effect preventif ethanol ectract of noni fruit to the decrease in the number of eritrocyte count in rats (Rattus norvegicus) which have been induced by methotrexate. The method used in this study is true experiment by using post test only control group design. The sample consists of 20 rats divides into 5 groups: (1) Group without any treatment (control); (2) Group which is induced of 0,54 mg/ day methotrexate (negative control);(3) Group which is given oral dose of 0,54 mg/ day methotrexate and 0,5 mg/ kg BW noni extract (treatment group 1); (4) Group which is given oral dose of 0,54 mg/ day methotrexate and 1 g/ kg BW noni extract (treatment group 2); (5) Group which is given oral dose of 0,54 mg/ day methotrexate and 2 g/ kg BW noni extract (treatment group 3). The eritrocyte is counted using blood chemistry analyzer and using statistical analyzes with Anova tes. The result from this study there is no effect showed between control, treatment 1, treatment 2, and treatment group 3 (P>0.05). Noni extract which is orally given have a tendency to increase the eritrocyte count of rats which have given oral dose of 0, 54 mg/ day methotrexate.

Abstrak: Mengkudu (Morinda citrifolia) merupakan tanaman yang sejak lama digunakan masyarakat sebagai pengobatan, salah satunya untuk pengobatan alternatif anemia yaitu dengan cara meningkatkan jumlah eritrosit. Hal ini dimungkinkan karena kandungan terpenoid pada mengkudu berfungsi untuk membantu pemulihan sel-sel tubuh. Penelitian ini bertujuan untuk mengetahui efek pencegahan ekstrak etanol buah mengkudu (Morinda citrifolia L) terhadap penurunan jumlah eritrosit pada tikus yang diinduksi metotreksat. Metode pada penelitian ini adalah eksperimental murni (True experiment) dengan menggunakan rancangan Post test only control group design. Sampel tediri dari 20 ekor tikus yang dibagi menjadi 5 kelompok : (1) Kelompok tanpa perlakuan (kontrol); (2). Kelompok yang diberi metotreksat 0,54 mg/hari (kontrol negatif);(3) Kelompok yang diberi metotreksat 0,54 mg/hari dan  ekstrak mengkudu 0,5 g/kgBB (perlakuan 1);(4) Kelompok yang diberikan metotreksat 0,54 mg/hari dan ekstrak mengkudu 1 g/kgBB (perlakuan 2); dan (5) Kelompok yang diberi metotreksat 0,54 mg/hari dan ekstrak mengkudu 2 g/kgBB (perlakuan 3). Perhitungan jumlah eritrosit  menggunakan Blood Chemistry Analyzer. Hasil uji statistik Anova menunjukkan tidak terdapat perbedaan yang bermakna antara kelompok kontrol, kelompok kontrol negatif, kelompok perlakuan, kelopok perlakuan 2, dan kelompok perlakuan 3 (p>0.05). Pemberian ekstrak etanol buah mengkudu secara oral memiliki kecenderungan untuk meningkatkan jumlah eritrosit tikus yang diinduksi oleh metotreksat.
