Pengaruh Pemberian Ekstrak Etanol Daun Jeruk Nipis (Citrus aurantifolia Swingle) Terhadap Penurunan Kadar Kolesterol Total Darah pada Mencit Model Hiperkolesterolemia

Abstract


Abstract: Hypercholesterolemia is a major risk factor of cardiovascular disease (CVD). Lowering total cholesterol level is one of main treatment strategy of CVD. Pharmacological treatment of CVD causes a side effects and financial burden on individuals and families. This study aimed to determine the effect of ethanol extract of citrus leaves (Citrus aurantifolia Swingle) on lowering total cholesterol level in hypercholesterolemic mice. This research was true experimental using random allocation sampling group design. Samples were 30 mice with swiss webster strain were divided into 5 groups: normal control, negative control, concentration 1.75g/kg, concentration 3.5g/kg, and concentration 7g/kg of ethanol extract of citrus leaves. Total cholesterol level measurements was done after the adaptation period, after 14 days were induced to be hypercholesterolemia, and after 14 days administration treated. Data were analyzed by Kruskal-Wallis test, the results shows a significant difference between the levels on lowering of total cholesterol after induction and after treated in all treatment groups after with p value = 0,002 (p<0,05). There was optimum dose at 3,5 g/kg between treatment groups The conclusion shows there were a significant difference effect of ethanol extract of citrus leaves on lowering total cholesterol level in hypercholesterolemic mice.

Abstrak: Hiperkolesterolemia merupakan faktor risiko utama penyakit kardiovaskular. Penurunan kolesterol total merupakan salah satu strategi dalam terapi penyakit kardiovaskular. Pengobatan secara farmakologis memicu terjadinya efek samping bagi penderita serta menyebabkan beban keuangan yang besar pada individu dan keluarga. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak ethanol daun jeruk nipis (Citrus aurantifolia Swingle) terhadap penurunan kadar kolesterol total darah pada mencit model hiperkolesterolemia. Metode penelitian ini adalah eksperimental murni dengan rancangan alokasi acak. Subjek penelitian adalah 30 ekor mencit galur swiss webster yang terbagi dalam 5 kelompok, yaitu kelompok kontrol normal, kontrol negatif, ekstrak ethanol daun jeruk nipis dengan konsentrasi 1,75g/kgBB, konsentrasi 3,5g/kgBB, dan konsentrasi 7g/kgBB. Pengukuran kadar kolesterol total darah dilakukan setelah masa adaptasi, setelah diinduksi hiperkolesterolemia, dan setelah 14 hari diberi perlakuan. Hasil uji statistik Kruskall-Wallis  menunjukkan pemberian ekstrak etanol daun jeruk nipis menurunkan kadar kolesterol total darah pada mencit model hiperkolesterolemia dengan nilai p=0,002 (p<0,05). Konsentrasi optimum pemberian ekstrak etanol daun jeruk nipis dalam menurunkan kadar kolesterol total adalah 3,5g/kgBB. Kesimpulan pada penelitian ini adalah ekstrak etanol daun jeruk nipis menurunkan kadar kolesterol total darah mencit model hiperkolesterolemia
