Efek Antiinflamasi Infusa Daun Pepaya (Carica papaya) terhadap Edema Telapak Kaki Tikus Jantan Galur Wistar yang Diinduksi Karagenin

Abstract


Abstract: Inflammation is a response of vascularized tissues to infections and damaged tissues that brings cells and molecules of host defense from the circulation to the sites where they are needed, in order to eliminate the offending agents. However, if excessive inflammation occurs, it can cause negative effect, which one is tissue damage. Inflammation process can be inhibitted by action of antiinflamatory agent. One of traditional plant which has antiinflammatory effect is papaya leaf. Papaya leaf has flavonoid which is  able to inhibit cyclooxygenase enzyme so it is able to inhibit prostaglandin forming. This study aims to determine antiinflammatory effect of papaya leaves infusion which edema induced in rat paw by carrageenan. The method used in this study were experimental with completely randomized design. This study used 30 white Wistar strain rats that divided into five groups: normal group, negative control group, positive control group (sodium diclofenac 2.25 mg/kgBB), infusion of papaya leaves 1st concentration (200 mg/kgBB), 2nd concentration (400 mg/kgBB), 3rd concentration (800 mg/kgBB). All of research subjects were adapted for seven days. Each research subject in each treatment group was given antiinflammatory agent then induced carrageenan so that creating an inflammatory reaction in the form of edema on the rat paw.  After that, change volume of paw edema before and after induction carrageenan measured using plethysmometer every hour for six hours continuously. The data obtained were statistically analyzed using Kruskal-Wallis and Mann-Whitney test. The results showed that papaya leaves infusion with 800 mg/kgBB has anti-inflammatory effect. Antiinflammatory effect of papaya leaves infusion due to flavonoid which is able to inhibit prostaglandin forming.

Abstrak: Inflamasi merupakan respon jaringan vaskular terhadap infeksi dan kerusakan jaringan yang membawa sejumlah molekul pertahanan host dari sirkulasi ke tempat yang diperlukan dan bertujuan untuk mengeliminasi agen yang mengganggu. Akan tetapi, jika inflamasi terjadi berlebihan dapat menyebabkan efek yang merugikan, diantaranya terjadi kerusakan jaringan. Proses terjadinya inflamasi dapat dihambat melalui kerja agen antiinflamasi. Salah satu tanaman obat tradisional yang memiliki efek antiinflamasi adalah daun pepaya. Daun pepaya mengandung flavonoid yang dapat menghambat enzim siklooksigenase sehingga dapat menghambat pembentukan prostaglandin. Penelitian ini bertujuan untuk mengetahui efek antiinflamasi infusa daun pepaya pada edema telapak kaki tikus yang diinduksi karagenin. Metode penelitian ini adalah eksperimental dengan rancangan acak lengkap. Penelitian ini menggunakan 30 ekor tikus galur Wistar yang terbagi dalam enam kelompok, yaitu kelompok normal, kelompok kontrol negatif, kelompok kontrol positif (natrium diklofenak 2,25 mg/kgBB), infusa daun pepaya konsentrasi I (200 mg/kgBB), konsentrasi II (400 mg/kgBB), konsentrasi III (800 mg/kgBB). Seluruh subjek penelitian diadaptasi selama tujuh hari. Setiap subjek penelitian pada masing-masing kelompok perlakuan diberikan agen antiinflamasi, kemudian subjek diinduksi karagenin pada telapak kaki agar timbul reaksi inflamasi berupa edema pada telapak kakinya. Setelah itu perubahan volume edema telapak kaki tikus sebelum dan sesudah induksi karagenin diukur dengan menggunakan plethysmometer setiap satu jam selama enam jam berturut-turut. Dikatakan memiliki efek antiinflamasi apabila didapatkan daya hambat inflamasinya > 50%. Data yang didapat dianalisis secara statistik menggunakan uji Kruskal-Wallis dan uji lanjut Mann-Whitney. Hasil penelitian menunjukkan bahwa infusa daun pepaya konsentrasi 800 mg/kgBB memiliki efek antiinflamasi. Efek antiinflamasi infusa daun pepaya disebabkan karena kandungan flavonoid yang dapat menghambat proses inflamasi dengan cara menghambat pembentukan prostaglandin.
